A vampire's humanity?

I was just wondering a few things about a vampire's humanity. Is it a voluntary thing oder do they eventually get desensitized and stop feeling? How many times did Stefan and Damon turn off their humanity? And how do Du know it was off? How about other Vampire on the show?
